Short answer: a defensible longevity protocol supports mechanisms known to decline with age — NAD+ availability, sirtuin activity, clearance of senescent cells — while being honest that no supplement has been shown to extend human lifespan. The behavioural inputs are not the garnish here; they are the main course.
Daily NAD+ foundation

The rationale for each component: NAD+ as supply, quercetin against degradation, resveratrol as downstream activation. Each of those is a mechanistic argument, and the strength of evidence behind them differs — which the next sections set out rather than blur.
NMN as the precursor
NMN, one capsule in the morning. NMN is the component of this protocol with the best human data: randomised trials have shown it raises blood NAD+ and, in specific populations, improves muscle insulin sensitivity and aerobic capacity. Whether to run NMN and NAD+ together or alternate is a budget question more than a physiological one — there is no trial comparing those schedules.
Quercetin and senescent cells
Quercetin has two proposed roles. It inhibits CD38, the enzyme that consumes NAD+ and whose activity rises with age — shown in knockout-mouse work by Camacho-Pereira and colleagues (2016). And it appears in the senolytics literature: Zhu and colleagues (2015) in Aging Cell identified quercetin, in combination with dasatinib, as selectively clearing senescent cells.
Both lines are preclinical, and the senolytic work used a drug combination rather than quercetin alone at supplement doses. This is a rationale for inclusion, not a demonstrated human outcome, and we are not going to describe it as one.
Resveratrol and the sirtuins
Sirtuins require NAD+ to function, which is the coherent half of the pairing. Resveratrol entered the field via Howitz and colleagues (2003) in Nature, which identified small-molecule sirtuin activators and reported lifespan extension — in yeast. Whether resveratrol activates SIRT1 directly in humans has been contested since.
An earlier version of this page cited a paper about folate transport as confirming resveratrol's sirtuin activation. That was simply the wrong paper, and the claim has been rewritten to match what is actually known.
The behavioural inputs — which are the bigger lever
Fasting and exercise both act on the same AMPK–NAD+–sirtuin axis these supplements target. Cantó and colleagues (2009) in Nature showed AMPK regulates energy expenditure by modulating NAD+ metabolism and SIRT1 activity — the mechanistic link between energy stress and this pathway.
Practically: regular aerobic and resistance training, adequate sleep, and not eating late are the primary drivers. A previous version of this page asserted that cold exposure activates SIRT3 in humans; we could not substantiate that and have removed it.

Realistic expectations
These supplements support cellular mechanisms associated with ageing. They have not been shown to slow or reverse human ageing, and no product should tell you otherwise. Chronic sleep deprivation, chronic stress and a poor diet each work against the same mechanisms faster than a supplement supports them.
