Short answer: magnesium supports the neurotransmitter systems that govern sleep onset and maintenance, and there is randomised human evidence that supplementation improves sleep measures in older adults with poor sleep. The glycinate form is preferred not because it has its own dedicated sleep trial, but because it absorbs well, is tolerated at a full nightly dose, and its glycine carrier has separately studied sleep effects.

Why magnesium matters for sleep

Magnesium is a cofactor in hundreds of enzymatic reactions, several of which govern the neurological conditions sleep requires. Intake is commonly below the recommended amount — the NIH Office of Dietary Supplements notes that a substantial proportion of people in the US consume less than the recommended intake from food.

Two mechanisms connect magnesium to sleep, and one supporting one.

GABA receptor function

GABA is the central nervous system's primary inhibitory neurotransmitter — the signal that quietens neural activity ahead of sleep. Magnesium supports GABA-A receptor function, so adequate magnesium status is a precondition for that braking system working normally. This is best understood as removing a constraint rather than as sedation.

NMDA receptor modulation

Magnesium sits in the NMDA receptor channel as a voltage-dependent block, damping excitatory glutamate signalling. Lower magnesium availability means a less effective block and a nervous system held at higher baseline arousal.

What the clinical evidence actually shows

The most-cited human trial is Abbasi and colleagues (2012) in the Journal of Research in Medical Sciences: a double-blind, placebo-controlled trial in 46 older adults with insomnia. Over eight weeks, supplementation improved insomnia severity index scores, sleep efficiency and sleep onset latency, and was accompanied by higher serum melatonin and lower serum cortisol.

One detail matters and is usually omitted: that trial used 500 mg of magnesium oxide, delivering roughly 250 mg elemental magnesium — not glycinate. An earlier version of this page presented Abbasi's findings twice, once attributed to a different journal and once to a magnesium bioavailability paper, as though they were two separate trials. They were the same study, and we have corrected that.

So the honest position is this: the randomised sleep evidence is for magnesium, in a poorly absorbed form, in older adults with insomnia. Why the glycinate form specifically

Magnesium glycinate is a chelate: the magnesium ion is bound to two glycine molecules, which shields it from phytates and oxalates in food that would otherwise reduce absorption. Comparative work such as Firoz and Graber (2001) established that magnesium oxide is very poorly absorbed relative to chelated and organic-acid forms.

The carrier is also not inert. In partially sleep-restricted volunteers, pre-sleep glycine improved subjective daytime performance and reduced fatigue in a trial in Frontiers in Neurology (2012). Preclinical work in Neuropsychopharmacology (2015) found glycine's sleep-promoting effect is accompanied by a fall in core body temperature, mediated by NMDA receptors in the suprachiasmatic nucleus — and a falling core temperature is one of the cues that precedes sleep onset.

That is two independent routes from one compound, which is the substantive reason to prefer this form for sleep.
